The leap from A up to high D, a fourth, just before the last note. The high D needs the most air in the tune, and then the B has to come in softer. On a 12-hole, Elegy of Emptiness goes from A to high D: you lift the left middle, left index and left thumb. On a 6-hole, you lift the left index and right thumb.

What does the Elegy of Emptiness do?
The song leaves "an empty shell of the form Link is in at the time", one for each of his four forms. Link needs the shells to climb the Stone Tower and to solve puzzles inside the Stone Tower Temple, and, as Zelda Wiki puts it, "shells can only be formed if Link is in any of the areas of Ikana Canyon".
